BV感染Hofbauer细胞的初步研究*
摘要 目的 探讨HBV感染Hofbauer细胞机制。方法 联合法、SP法分离鉴定人绒毛Hofbauer细胞;ELISA法检测培养液上清HBVM,PCR法检测Hofbauer细胞及培养上清中HBV-DNA。结果 使用联合法可获取高活度、高纯度的Hofbauer细胞;HBV感染血清可无辅剂下直接依附Hofbauer细胞表面,并可侵入细胞内进行复制表达。结论 HBV感染血清可直接感染Hofbauer细胞。

Preliminary study on infection of HBV in Hofbauer cells of human villus in vitro

[Abstract] Objective To explore the molecular mechanism of HBV infection in Hofbauer cells of human villus. Methods Hofbauer cells in human villus were isolated and purificated by the union separation and identificated by the method of streptavidin-peroxidase (SP) immunocytochemical. HBVM and HBV-DNA in supernatant and Hofbauer cells were detected by ELISA and PCR, respectively. Results The high purity and activity of Hofbauer cells could be obtained by the union separation. Hofbauer cells could be infected by HBV positive serum whitout other assistant and some viral particle of HBVcould be released. Conclusion Cultured Hofbauer cells can be directly infected by HBV positive serum in vitro.

目前全世界HBV感染者约20亿,HBV慢性携带者约3.5亿,其中40%~50%是由母婴垂直传播所致[1]。在介导HBV宫内感染过程中,胎盘起重要作用。正常情况下,母血和脐血分别在各自封闭的管道内循环,互不相混,两者之间隔有胎盘屏障,胎盘屏障从母面到子面依次是滋养层细胞、绒毛间质细胞和毛细血管内皮细胞。母血HBV要感染胎儿须先穿透胎盘屏障。目前,国内外学者对HBV体内外感染滋养层细胞做了部分研究[2],但滋养层细胞靠近母血,若要感染胎儿则须进一步感染绒毛间质细胞及毛细血管内皮细胞。Hofbauer细胞是胎盘的巨噬细胞,位于绒毛间质,毗邻母血的滋养层细胞和靠近脐血的绒毛毛细血管细胞。为探讨Hofbauer细胞在HBV宫内感染具体机制,本文进行HBV体外感染Hofbauer细胞的初步研究,现将结果报告如下。
